Onco-Innovations Advances ONC010 Toward Clinical Trials with High-Purity Manufacturing and Australian Pathway

Onco-Innovations Limited (CBOE CA: ONCO) (OTCQB: ONNVF) (Frankfurt: W1H) has announced a major advancement in the development of its lead candidate ONC010, a small-molecule PNKP inhibitor designed to disrupt DNA damage repair in cancer cells. The company has successfully completed key manufacturing and preclinical steps, positioning ONC010 for clinical evaluation.

The company reported that a 300-gram development batch of ONC010's active ingredient achieved approximately 99.3% purity, a critical milestone for pharmaceutical-grade production. Additionally, Onco-Innovations identified a non-tin catalyst that could improve impurity control in future polymer manufacturing, enhancing the scalability and quality of the nanoparticle delivery system.

ONC010 targets Polynucleotide Kinase Phosphatase (PNKP), an enzyme involved in the DNA damage response (DDR) pathway. By inhibiting PNKP, ONC010 aims to impair cancer cells' ability to repair DNA damage, making them more susceptible to treatments like chemotherapy and radiation. This approach addresses a fundamental challenge in oncology, where cancer cells often resist therapy by repairing the damage inflicted by DNA-damaging agents.

The company is building a regulatory pathway in Australia for a potential First-in-Human study, with ongoing work in bioanalytical, metabolism, ADME, toxicology, and biodistribution studies scheduled for the second half of 2026. These efforts are part of a comprehensive preclinical program designed to meet regulatory requirements and ensure the safety and efficacy of ONC010 before human trials begin.

"We are excited about the progress we have made with ONC010," said a company spokesperson. "The high purity achieved in manufacturing and the identification of a more efficient catalyst underscore our commitment to developing a robust and scalable production process. Our focus now is on completing the necessary preclinical studies to advance ONC010 into clinical trials, with the goal of providing a new therapeutic option for cancer patients."

The latest update from Onco-Innovations highlights the company's strategic approach to drug development, leveraging its proprietary nanoparticle technology to enhance the delivery and efficacy of PNKP inhibitors. As DDR inhibitors gain traction in oncology, ONC010's unique mechanism and formulation could position it as a promising candidate in the fight against cancer.
